Hamlin started his football career in the Catholic High School located in Pittsburgh. A decent form throughout his high school career earned him the All-State and the Class AAAA Defensive Player of the Year awards. His form was noticed by many colleges and they were trying to get Hamlin into their colleges. But Damar wished to continue his career with Pittsburgh university. During his freshman year, he was injured and redshirted, he just played three matches in the season. In his sophomore season, he performed 76 tackles and two interceptions as a redshirted sophomore. He was named the All-Atlantic Coast Conference at the end of his sophomore season. His form continued in both his junior and final years.

After his brilliant performance in his college career, he entered the NFL draft with lots of expectations. As he expected, he was drafted by the Buffalo Bills as the 212th overall pick in the 6th round of the 2021 NFL draft. As a rookie, he signed a pretty good contract.

On January 2 2023, in a match against Cincinnati Bengals, Damar collapsed while trying to tackle the Bengals Wide Receiver, Tee Higgins. He has completely left out of consciousness and a team of doctors rushed into the field. Immediately after checking on him, he was taken on a stretcher in the ambulance to the University of Cincinnati Academic Health Center. The hospital released a health report stating that he suffered from a cardiac arrest and was in critical condition. This left his teammates and the NFL community in great shock.
